You check into the hotel and with your room key, you go to a kiosk in the hotel to print out the express pass. That is separate from actually having park tickets. The length of your hotel stay dictates how long you can use the pass.

Yes, the desk clerk will give you a temporary room key (that doesn't open any room) to use to get the express pass card. And if checking in early and you are club level, you can get access to the club room for breakfast even though your room is not yet ready.
